Hello,
Is it possible to join Skype for business meetings from a standalone Cisco video endpoint (ex: MX300 or Room55) ?
Does it require additional infrastructure? What about licensing ?

You will need additional infrastructure to enable you to connect proper Standards Based Video Conferencing Endpoints with Skype for Business. This could be any or all of the following:- Cisco Meeting Server, Cisco Webex, Cisco VCS with Microsoft Interoperability Option, Pexip Infinity. Any of these can do the translation from one environment to the other if configured correctly. The two cannot direcly interface together.

If you only have a small number of TelePresence endpoints, then the "Cisco VCS with Microsoft Interoperability Option" is probably not the best answer as it is extremely expensive.

Currently only 1 Telepresence is required. Liek you said the VCS is expensive for such a small deployment.

None of the options are "easy". Each one has its differences, limitations and challenges. None are particularly easier than others.
It gets even worse if you need to talk to Microsoft Teams - so avoid that if at all possible at this point in time (unless you pick the Pexip solution).
