Hey Kyle,
Seems like you are running into CDET http://tools.cisco.com/Support/BugToolKit/search/getBugDetails.do?method=fetchBugDetails&bugId=CSCtg93965
As per the CDET:
If Click To Call setting was changed from "Preferences", user doesn't click on the 'Phones' tab but instead clicks on the 'Save' button, the setting does not get changed.
The workaround is to click on the 'Phones' tab and then save.
There is an ES available which has the fix for this BUG. To obtain that ES, please open a TAC Service Request.
If the issue still persists after following the workaround, then we need to see if there is UAC or Admin Rights Issue on the Local PC.

It looks good, I was able to save the setting, however whenever I initiate a call I receive an error message
Unexpected end of file, no input , or timeout while receiving data, please contact your administrator
thx for all of your help

How did you manage to fix this issue ? It's not working for me.I am able to save the config, but when I initiate the call I got the same message as you (unexpected end of file ....)
BTW, which version of MS Office/ Outlook are you using ?
Thanks

Hi These are OS level support and doesnt really confirm if you can use it within any appliucation installed natively on 64bit. I would like to use click to call for dialing contacts in my outlook 2010. thsi works from 32bit office 2010 but not 64bit

I got your point buddy!
Click to Call only supports 32-bit versions of Microsoft Office 2010 applications.
