Yes, it is quite doable.  Depending upon what operating system you plan to boot, the process will obviously vary.  What OS do you want to boot?

This document - Cisco Virtualization Solution for EMC VSPEX with VMware vSphere 5.1 for 100-125 Virtual Machines - Cisco - contains information on how to set it up.  I believe it is using 10 GE, but the process is exactly the same.  It contains a lot of other information, too, but it does give instructions for setting up iSCSI boot for ESXi 5.1.  Should be the same for 5.5.
