Hi,
Would it be possible to config Box-to-Box HA on ISR 4321 router ?. ISR4321 comes with 2 GE interfaces while the configuration guide example with 3 interfaces.

Got your point, looks like 3 ports are required for ISR 4K to achieve box to box.
ISR4321comes with 2 ports, and all config guides references are for 3 ports.
I think open up a case with Cisco is your best bet.

As @Ratheesh Kumar said, it will not be possible to configure basis the design guidelines. If you think of configuring LAN and WAN as sub-interfaces on single interface then you need to validate with Cisco TAC for design supported. I dont see that is validated design.
As best approach I would recommend is to procure the GLC 1 GB copper module and connect into SFP module. As per ISR 4000 series sub ISR 4321 specs, there is feasibility to introduce one more copper module. Please check the rear view of SFP port where the Gig Port -3 would be mentioned.
