We have a number of clients who would be keen of UCCX email routing if Office 365 were supported. Is an option at the moment to use a Hybrid deployment, office 365 with on prem exchange for only the mailboxes we want to integrate to UCCX agent email?

There is nothing as such that Office 365 lacks something, infact it is one of the best suite for Exchange available at the moment. However, it is about the testing of it and other compatibility stuff that had not been tested yet with UCCX.
