I have around 4000 users is it needs license?

BAT is the way and with your version you will need 1 DLU per mobility enabled user.

After I used BAT, is I need to change the "Destination Number" for each user one by one? - Or you can change it from Remote destination csv file - "DESTINATION" box in the RD csv.
